The ImageTrak is an innovative system that integrates fiber optic epi-fluorescence imaging with PlateTrak automated liquid handling robotics to provide a high capacity, high throughput system for assay miniaturization and lead optimization. The ImageTrak is unique in its ability to combine proven Packard automation, sensitive imaging techniques and data analysis features into one complete system for high throughput screening. Two ImageTrak models are available to handle varied assay requirements. Each system is equipped with 96-channel dispensing for automated processing of 96 to 1536-well high-density microplates, a bi-directional conveyor for secure plate handling and an epi-fluorescence imaging module in a light tight enclosure. Based on patented “contact imaging” technology, the ImageTrak uses fiber optics and a cooled CCD camera to achieve a high level of sensitivity, enabling this integrated system to handle many different assays. The ImageTrak utilizes 96 individual fiber optic bundles with separate optical paths for excitation and emission light to enable simultaneous measurement of 96-wells in dual wavelengths for true ratiometric FRET and kinetic assays. The ImageTrak’s optical system is mounted from below and built into Packard’s unique ultiPosition Dispense (MPD) module design. The MPD combines Dispense Head movement, tip and plate handling, and liquid dispensing into a single unit for precise automation control. Using the MPD, the ImageTrak can accurately position microplates on the contact imager with tips loaded, enabling simultaneous “dispense and read” capabilities. The ImageTrak HT model offers an additional 10-inch MPD with 384-channel Dispense Head to pre-load dyes and dispense compounds for high throughput liquid handling. APPLICATIONS FOR AUTOMATED ASSAY SCREENING Plate washing, dispensing, dye loading, timed incubations and compound, agonist or antagonist addition. Image acquisition protocols including linked dispense-and-read sequences. Applications include calcium assays, membrane potential, FRET assays for ion-channels and quencher assays, and prompt fluorescence. The Imaging Detector software module allows the user to select key assay parameters including excitation and emission filters, integration time, plate type and dispense timing. Acquisition modes include single point readings and kinetic sequences, with variable intervals. Kinetic sequences include repeat readings and cycling readings between sub-wells (for 384- or 1536- well plates). Assay protocol files can be stored with parameter settings and controls for defined assays. Image acquisition times variable from 0.5 s to 30 s, typically 1s readings. sequences are linked to reagent dispense functions. This allows the user to define read sequences with baseline readings for one or more dispense-and-read steps for cell-based assays. Data output is available in list format or plate format, for single wavelength, dual wavelength or ratiometric data. A user configurable report file output is available. DISPENSE AND READ Baseline, injection and read SENSITIVITY Fluorescein sensitivity in the range 5 x 10 -11 M, in 96-well and 384-well plates. Sensitivity depends on specific assay conditions.
